Bridging CRISPR biology and technologies

The term CRISPR has become synonymous with genome editing, whether for reversing genetic diseases in humans, enhancing crop plants, or achieving genetic pest management. It has also generated ample controversy with the recent announcement of CRISPR babies and an ensuing moratorium on germline editing in the clinic. However, CRISPR was not invented in a laboratory but instead discovered as the basis of adaptive immune systems in bacteria and archaea. These systems employ RNA guides to direct the system’s protein machinery tomatching DNA or RNA sequences from foreign invaders, thereby cleaving these sequences and blocking the invasion. The original insights into these CRISPR-Cas systems directly lent to many of the CRISPR technologies that have revolutionized genome editing. At the same time, CRISPR-Cas systems have proven incredibly diverse, with new proteins, mechanisms, and functions still being discovered. My group has endeavored to uncover the functional diversity of CRISPR-Cas systems and translate insights into new tools and technologies. As part of this talk, I will discuss some of our ongoing work at the intersection between CRISPR biology and technologies, including elucidating rules for target recognition as well as constructing and implementing CRISPR arrays. Through these examples, I will illustrate how much we have to learn about CRISPR as adaptive immune systems and how new findings can contribute to the already disruptive use of CRISPR technologies for genome editing and beyond.
